Expression of p53 and nm23-H1 Protein in Laryngeal Squamous Cell Carcinoma

ABSTRACT

P53 tumor suppressor gene has come to the forefront cancer research, because it is frequently mutated in human cancers. And nm23-H1 gene product is known as a mediator of cancer cell invasion and metastasis. But there are still controversy in their clinical meanings in head and neck cancer, especially in laryngeal squamous cell carcinoma. Immunohistochemical stainings with monoclonal antibody against p53(NCL-p53-DO7) and nm23-H1 (NCL-nm23) was done to investigate their clinical meanings in laryngeal squamous cell carcinoma. The results are as follows : 1) There was no significant difference in the positive rates of p53 and nm23-H1 expression between glottic and supraglottic squamous cell carcinoma. 2) The positive rates of p53 expression significantly increased with T-stage but was not affected by N-stage and histopathologic grades. 3) The negative rates of nm23-H1 expression significantly increased with T-stage and decreased as neck metastasis developed. In conclusion, there may be no cell biologic difference between glottic and supraglottic squamous cell carcinoma. But mutant p53 protein can be a biologic marker for primary tumor progression, and loss of nm23-H1 protein can be a worse prognostic marker for tumor invasion and metsatasis.
