This study was performed in order to identify the glycoproteins containing oligosaccharides in adult rat soft palate. After making serial paraffin sections at 4μm, the specimens were incubated with 9 species of lectins(UEA-I, BS-I, DBA, SBA, WGA, PNA, PNA after neuraminidase treated, Con-A, RCA-I, LCA). In order to increase specificity of reactions, the sections were applicated with ABC system. And then the sections were incubated DAB and were counterstained with hematoxylin. The other sections were also stained with hematoxin and eosin. The results summarized were as follow ; 1) In the epithelium : a) The coats of cell membranes in the granular cell layer were stained with SBA. b) The coats of cell membranes in the upper spinous cell layer were stained with UEA-I, SBA, WGA, Con-A and RCA-I. c) The coats of cell membranes in the lower spinous cell layer were stained with UEA-I, BS-I, SBA, WGA, PNA, PNA after neuraminidase treated, Con-A and RCA-I. d) The coats of cell membranes in the basal cell layer were stained with BS-I, WGA, PNA after neuraminidase treated Con-A, RCA-I and LCA. 2) In the basement membrane, all lectins used were not stained. 3) In the lamina propria, the connective tissue matrix was stained with UEA-I, BS-I, DBA, WGA, PNA after neuraminidase, Con-A, RCA-I and LCA. 4) In the mucous gland, the mucous cell were stained with UEA-I, DBA, WGA, RCA-I and LCA.
