ABSTRACT |
Diethyldithiocarbamate(DDTC) has been reported to protect against renal toxicity of cisdiamminedichloroplatinum(cisplatin) in experimental study, but the mechanism of action of renal protection and the effects on cell kinetics of DDTC and cisplatin to the normal organs were not reported. In this study, change of the labeling index of basal cells of tongue epithelium of the mice by demonstration of bromodeoxyuridine(BrdU) labeled S-phase cells using immunohistochemical methods with anti-BrdU, antibody and blood urea nitrogen(BUN) value and serum creatinine level were analyzed in mice after intraperitoneal injection of cisplatin with or without DDTC. The labeling index of the basal cells of tongue epithelium increased in ascending orders in group treated with 12mg/kg cisplatin, group with 6mg/kg cisplatin, group with 12mg/kg cisplatin in combination with DDTC 1mg/kg and control group. The blood urea nitrogen value decreased in descending orders in group treated with 12mg/kg cisplatin, group with 12mg/kg cisplatin in combination with DDTC 1g/kg, group with 6mg/kg cisplatin and control group. The serum creatinine levels have no statistical correlations among the each groups. This results suggested that DDTC protects the nephrotoxicity of cisplatin and inhibits cisplatin effects of decreasing labeling index of basal cells of tongue epithelium in normal mice.
